Embracing Cancer Complexity: From Drosophila Avatars to the Chemputer

Abstract
Cancer clinical trials have among the lowest success rates of any major disease area. A central barrier is cancer’s whole-body complexity. I will describe our work developing network-based models and therapeutics designed to address this complexity, including broad mechanisms of drug resistance. Using colorectal cancer as a case study from our fly-to-bedside clinical trial, I will show how personalised fly avatars can identify novel, bespoke drug cocktails. I will then describe our work with Lee Cronin and colleagues, combining chemical evolution with digital chemistry to automate the targeting of complex biological networks.
Together, phenotype-led models and automated chemistry offer a new path towards cancer therapies built for whole-body complexity.
